<p>To improve our understanding of the genetic diversity and population structure of <i>Metapenaeus intermedius</i> for germplasm resource conservation, we developed and characterized 67 polymorphic microsatellite markers for this species. The number of alleles per locus ranged from 3 to 17. The observed heterozygosity (<i>H</i><sub><i>O</i></sub>) varied from 0.2854 to 0.9875, whereas the expected heterozygosity (<i>H</i><sub><i>E</i></sub>) ranged from 0.2957 to 0.9327. Twenty-nine loci exhibited moderate polymorphism (0.25 &lt; <i>PIC</i> &lt; 0.5), while the remaining loci demonstrated high polymorphism (<i>PIC</i> &gt; 0.5). Furthermore, 10 loci deviated from the Hardy‒Weinberg equilibrium (<i>P</i><sub><i>HW</i></sub>). These markers will serve as valuable tools for population genetic analyses and the conservation of <i>M. intermedius</i>.</p>
