Background <p>Previous studies have suggested a positive causal relationship between schizophrenia and ovarian, breast, and lung cancers. Additionally, research has indicated a positive correlation between bipolar disorder and thyroid cancer. However, some studies have also shown that mental disorders do not increase the risk of cancer, however, the causal relationship has yet to be thoroughly explored. In this study, we employed Mendelian randomization (MR) to examine the genetic connections between mental disorders and the risk of gynecological cancers.</p> Methods <p>We analyzed data from genome-wide association studies (GWAS) on five mental disorders [schizophrenia, bipolar disorder (BD), major depressive disorder (MDD), attention-deficit hyperactivity disorder (ADHD), and autism spectrum disorder (ASD)] and three gynecological cancers [cervical cancer (CC), ovarian cancer (OC), and endometrial cancer (EC)]. Instrumental variables were identified using significant single nucleotide polymorphisms (SNPs). We applied inverse variance weighting (IVW), weighted median (WM), and MR-Egger regression methods to assess the results, reporting effect sizes as odds ratios (OR) with 95% confidence intervals (CI).</p> Results <p>Genetically predicted MDD was found to be causally associated with an increased risk of LGSOC (OR = 1.50; 95% CI: 1.07–2.09; <i>P</i> = 0.01), MOC (OR = 1.46; 95% CI: 1.0–1.94; <i>P</i> = 0.008), BD with a decreased risk of EEC (OR = 0.76, 95% CI: 0.60–0.97, <i>P</i> = 0 .0249), and ADHD with an increased risk of LGSOC (OR = 1.48, 95% CI: 1.04–2.1, <i>P</i> = 0.0293).</p> Conclusions <p>Our study provides evidence of a causal relationship between genetically predicted MDD and OC risk, as well as between ADHD and OC risk. Additionally, we found that BD is linked to a reduced risk of EC.</p>

The causal association between psychiatric disorders and gynecological cancer: a bidirectional Mendelian randomization study
