Unveiling the Role of Mesalamine-Loaded Nanocarriers for Advanced Targeted Cancer Treatment

5-Aminosalicylic acid or mesalamine is an anti-inflammatory nonsteroidal drug considered a safe and effective medication, and side effects are uncommon. Recent advancements in nanoparticle systems are improving the delivery of mesalamine to inflamed areas, enhancing its solubility and bioavailability while minimizing side effects. The review examines the utilization of various nanocarriers, including lipid-based, polymeric, inorganic nanostructures, and carbon-based, to increase mesalamine’s bioavailability, solubility, and permeability while minimizing side effects. It highlights contemporary nanoparticle drug delivery systems (DDSs) developed explicitly for targeted delivery to the colon, focusing on treating colorectal cancer and colon inflammation. Polymeric nanoparticles are being investigated for targeted colon delivery, preventing premature absorption. Stimuli-responsive nanoparticles that react to specific conditions, such as pH, ensure mesalamine’s release in the colon. Lipid nanoparticles enhance the targeting of inflammatory bowel diseases, resulting in lower dosages and side effects. Inorganic nanoparticles provide stability and functionalization benefits, while carbon-based nanoparticles protect mesalamine from degradation, facilitating efficient delivery to target sites.
