Tracking Surgical Impact Through Salivary Exosomes in Oral Squamous Cell Carcinoma: An Original Research

Oral squamous cell carcinoma (OSCC) remains a major global health burden due to high morbidity and mortality associated with late-stage diagnosis. Salivary exosomes have emerged as promising non-invasive biomarkers for cancer detection and treatment monitoring.
AimTo evaluate quantitative and qualitative changes in salivary exosomes in OSCC patients before and after surgical intervention.
Materials and MethodsIn a prospective clinical study, saliva samples were collected from OSCC patients pre-operatively and at defined post-operative intervals. Exosomes were isolated and analyzed using Nanoparticle Tracking Analysis (NTA) for concentration and size distribution, and Zetasizer analysis for zeta potential measurements. Statistical comparisons were performed using appropriate parametric and non-parametric tests.
ResultsPre-operative samples showed heterogeneous exosome populations with a peak around 100 nm, while post-operative samples displayed a more uniform distribution and reduced small vesicles. A significant decrease in total exosome concentration and a marked shift in zeta potential were observed post-surgery (p < 0.05), indicating reduced tumor-derived exosomes and altered exosomal composition.
ConclusionSalivary exosomes exhibit measurable changes following surgical treatment for OSCC, supporting their role as non-invasive biomarkers for tumor burden, treatment response, and recurrence monitoring. Corroboration-based management for OSCC patients must be verified by evidence-based scientific research.
