<p>The efficient degradation of lignocellulosic biomass is essential for the sustainable production of industrially relevant enzymes such as cellulases. Rumen microorganisms are recognized as promising candidates for cellulase production due to their ability to thrive under anaerobic conditions and utilize complex plant-based substrates. In this study, cellulase-producing bacteria were isolated from rumen samples, and the EB7 isolate was identified as the most potent producer, exhibiting an initial enzyme activity of 12.4 ± 0.42 U/mL. Molecular characterization based on 16&#xa0;S rRNA gene sequencing confirmed that the isolate belongs to <i>Streptomyces violaceochromogenes</i>. A systematic optimization of physicochemical and nutritional parameters was then performed to enhance cellulase yield. The optimal conditions pH 9, temperature 37&#xa0;°C, incubation period 3 days, 2&#xa0;g/L yeast extract, and 30&#xa0;g/L watermelon peel powder (WPP) resulted in a marked improvement in enzyme productivity, reaching 78.9 ± 3.78 U/mL, corresponding to an approximately six-fold increase compared with unoptimized conditions. These findings demonstrate the strong potential of rumen-derived <i>Streptomyces</i> strains for sustainable enzyme production and highlight the value of agro-waste valorization as a cost-effective and eco-friendly approach to industrial bioprocess development.</p>
