<p>In this study, our goal is to determine the chemical composition, to quantify and identify the phenolic compounds by UPLC-ESI-MS/MS, and to evaluate their antioxidant effect by proceeding with several assays: DPPH, ABTS, β-carotene bleaching, reducing power, O-phenanthroline, total antioxidant capacity (TAC) and silver nanoparticle of <i>Opuntia fcus indica</i> (OFI) seeds. The sunscreen ability against UV radiation (photoprotective effect), DNA damage protection effect, and inhibitory activities of tyrosinase, acetylcholinesterase (AChE), and butyrylcholinesterase (BChE) enzymes by Algerian OFI seeds. The finding demonstrated that OFI seeds are rich in carbohydrates (66.59 ± 1.79%) and contain notable quantities of proteins (6.12 ± 0.6%), fats (7.80 ± 0.03%), and ash (1.20 ± 0.06%), respectively. The seed extracts also had an appreciable content of polyphenols, ranging from 129.61 to 149.42&#xa0;µg GAE/mg. The UPLC-ESI-MS/MS analysis identified several phenolic compounds, including quercetin, rutin, and gallic acid. The results of antioxidant activities showed different values ranging from 41.72 to 97.55&#xa0;µg/mL for IC<sub>50</sub> results and from 28.01 to 165.83&#xa0;µg/mL for A<sub>0.5</sub> results. The extracts exhibited both photoprotective and DNA damage protection effects. Additionally, good IC<sub>50</sub> values were recorded in inhibitory activities of tyrosinase and cholinesterase enzymes, ranging from 86.93 to 301.45&#xa0;µg/mL for OFI seed extracts. The results of this work have shown that the phenolic extracts of OFI could constitute a good choice for nutrition, pharmaceuticals, and cosmetic applications.</p>
