Purpose of Review <p>Cyclin-dependent kinase (CDK) 4 and 6 inhibitors have revolutionized treatment for patients with hormone receptor-positive (HR +) breast cancer but have significant associated adverse events. This review provides an updated summary of the existing literature regarding the adverse event profiles for CDK 4/6 inhibitors. We also discuss supportive care recommendations for patients experiencing CDK-related toxicities.</p> Review Findings <p>Adverse events related to CDK 4/6 inhibitors occur across a broad spectrum, including hematologic, gastrointestinal, cutaneous, cardiac, and respiratory toxicities. Non-pharmacologic supportive care interventions to consider for patients with HR + breast cancer taking CDK 4/6 inhibitors include cognitive behavioral therapy strategies such as psychoeducation, relaxation exercises, mindfulness, patient assertiveness training, and hypnosis.</p> Summary <p>Patients with HR + breast cancer taking CDK 4/6 inhibitors are likely to experience adverse events that may impact their quality of life. Integrating supportive care strategies to mitigate CDK 4/6-related adverse events may be beneficial for patients in addition to clinical management strategies.</p>
