<p>Antibiotic-associated diarrhea (AAD) is an adverse reaction caused by antibiotic treatment, typically resulting from an imbalance in the gut microbiota. Probiotics have been widely used to counteract AAD by enhancing the composition of intestinal microbiota. Canine-derived <i>Pediococcus acidilactic</i> GLP06 has been shown to be a promising probiotic, however, the effect of <i>P. acidilactici</i> GLP06 on canine AAD is unknown. In this experiment, we investigated the effect of <i>P. acidilactici</i> GLP06 on antibiotic-induced diarrhea induced by intestinal flora imbalance in Beagle dogs by blood biochemical index test combined with 16s rRNA sequencing, SCFA test, and fecal untargeted metabolomics analysis. The results showed that probiotic administration significantly improved faecal scores, reduced serum diamine oxidase and tumour necrosis factor-α levels (<i>p</i> &lt; 0.05), improved the intestinal barrier and inhibited intestinal inflammation. Importantly, after treatment with <i>P. acidilactici</i> GLP06, gut microbial diversity and richness gradually recovered, with a significantly increase in the number of <i>Lactobacillus</i>, <i>Bifidobacterium</i>, and <i>Lachnospirales</i> that produce short-chain fatty acids (SCFAs) (<i>p</i> &lt; 0.05), and promoted the production of acetic acid, butyric acid, and total SCFAs in the gut (<i>p</i> &lt; 0.05). In addition, metabolomics analysis showed that <i>P. acidilactici</i> GLP06 reversed the interference of antibiotics on major metabolic pathways such as purine metabolism, sphingolipid signaling pathway and bile secretion and increased the production of SCFAs, unsaturated fatty acids and glycocholic acid and other beneficial metabolites. These findings indicate that <i>P. acidilactici</i> GLP06 reversed the rupture of antibiotics on canine intestinal health by improving intestinal barrier function, regulating intestinal microbiota and restoring metabolic homeostasis, and effectively alleviated canine AAD. This provides scientific evidence for the potential use of <i>P. acidilactici</i> GLP06 as a probiotic for the prevention and treatment of canine AAD in future clinical applications.</p>
