Natural Antimicrobial Preparation Source of N+ p Multivoltine Silkworm, Bombyx mori L.: Efficacy Against Various Bacterial Strains

Nistari, an indigenous multivoltine silkworm breed, is valued in Eastern India for its climatic resilience and pathogen resistance. The haemolymph of Bombyx mori L. larvae, particularly Nistari (N + p), contains antimicrobial peptides (AMPs) crucial to innate immunity. In this study, haemolymph-derived peptides were purified and tested against pathogenic bacteria. Crude haemolymph (13.97 mg/ml protein) showed strong activity against Escherichia coli, Salmonella typhi, Staphylococcus aureus, and Pseudomonas aeruginosa. Reverse-phase high performance liquid chromatography (RP-HPLC) isolated five fractions (SP1–SP5), with SP2 and SP4 showing superior broad-spectrum activity, exceeding commercial antibiotics. SP4 was most potent, producing inhibition zones of 28.16 mm against S. typhi and 28 mm against S. aureus. Matrix-assisted laser desorption/ionization (MALDI) and liquid chromatography-electrospray ionization-mass spectrometry (LC-ESI-MS) analyses identified SP2 (17 residues, 2088 Da) and SP4 (33 residues, 3399 Da) as leucine-rich, amphipathic peptides with α-helical structures that disrupt microbial membranes. These results suggest B. mori L. haemolymph peptides as promising alternatives to conventional antibiotics for combating resistance.
