Artificial masculinization of underyearling leopard coral grouper Plectropomus leopardus through oral administration of 17α-methyltestosterone

The leopard coral grouper Plectropomus leopardus is a valuable species in aquaculture, particularly in China and Southeast Asia. However, as a protogynous hermaphrodite, the time required to develop male broodstock remains a challenge for efficient aquaculture of this species. In this study, masculinization was artificially induced in underyearling P. leopardus through oral administration of 17α-methyltestosterone (MT) during gonadal sex differentiation to enhance the efficiency of grouper aquaculture and breeding. MT administration from 63 to 180 days after hatching, at a concentration of 50 μg/g of feed, effectively induced masculinization and spermatogenesis. However, testicular cells, including sperm, disappeared within 1 month after MT withdrawal, which may indicate the importance of continuous MT administration to maintain masculinization. In addition, a small number of fertilized eggs and hatched larvae were obtained through artificial fertilization using intragonadal sperm from MT-treated fish. These findings indicate that MT can induce functional sperm in underyearling P. leopardus, thereby shortening the generation time.
