<p>CD44, a transmembrane glycoprotein involved in cell adhesion, migration, and signaling, plays a complex role in cancer biology. This study aimed to investigate the expression patterns and molecular correlations of CD44 in human breast cancer tissues compared to benign fibroadenoma samples. Using qRT-PCR and immunohistochemistry, we found that CD44 gene expression was upregulated in 62.5% of breast cancer cases. Notably, CD44 expression showed significant positive correlations with BRCA1, p38 MAPK, E-cadherin, DSG1, HOTAIR, and BC200, suggesting a role in both stemness and tumor progression pathways. However, no correlation was observed with AKT, HER2, ERα, or endocytosis-related genes such as clathrin and caveolin-1. Immunohistochemical analysis revealed cytoplasmic and nuclear CD44 localization in malignant tissues, with stronger nuclear staining observed in higher tumor grades. These findings highlight CD44’s multifaceted role in breast cancer, supporting its involvement in tumor progression while emphasizing the need for further research to clarify its context-dependent functions.</p>
