<p>We present a robust method for global DNA methylation analysis based on complete acid hydrolysis and mass spectrometry. Unlike enzymatic approaches, it is independent of sequence context and shows high reproducibility. Application to sea lettuce <i>Ulva</i> revealed microbiome-dependent changes in DNA methylation, highlighting its potential for quantitative epigenetic studies.</p>
