Antisense oligomers (ASOs) turn sequence information into programmable anti-infective agents and microbial precision tools. Short peptide nucleic acids (PNA), combined with delivery carriers, enable targeted manipulation of microbial translation, including blockade of essential genes as asobiotics. Here, we summarize mode of actions, recent advances, and remaining gaps in the design rules for target selection, delivery, and resistance.
