The gut microbiome plays a crucial role in human health, but the molecular mechanisms underlying microbial interactions remain poorly understood. Activity-based protein profiling (ABPP) enables functional annotation of uncharacterized enzymes, particularly proteases, using chemical tools. Applying ABPP, Bacteroides fragilis Protease 1 (Bfp1) was identified as an activator of the human protease-activated receptor 2 (PAR2), modulating intestinal inflammation and barrier integrity.
