<p>Purpose: Endophytic fungi can synthesize plant secondary metabolites, offering new avenues for medical research. They are being screened for valuable compounds like taxol, camptothecin, and podophyllotoxin. Vinblastine and vincristine are excellent anti-cancer drugs but their current production using plants is non abundant and expensive. Methods: Current study focused on isolation of endophytic fungi from <i>Catharanthus roseus</i> (L.) G. Don plant collected from Bangalore, Karnataka, India. In the present study endophytes isolated from leaves of <i>Catharanthus roseus</i> were screened for vinblastine by specific biochemical tests and tryptophan decarboxylase encoding gene (TDC). A fungal isolate exhibiting the presence of the TDC gene was identified as <i>Colletotrichum siamense</i> through molecular identification of the internal transcribed spacer (ITS) sequence. Results: The presence of vinblastine was quantified by high-performance liquid chromatography (HPLC), yielding 138&#xa0;µg/L. Its structure was confirmed by electrospray ionization mass spectrometry (ESI-MS) with a molecular ion at <i>m/z</i> 811, ultraviolet (UV) spectroscopy showing absorption at 220–254&#xa0;nm, and Fourier-transform infrared (FTIR) spectroscopy identifying characteristic functional groups. The cytotoxic activity of the partially purified fungal vinblastine was evaluated using MTT assay on human cervical carcinoma (HeLa) and breast adenocarcinoma (MCF- 7) cell lines. The IC<sub>50</sub> values were found to be 23.89&#xa0;µg/mL and 19.9&#xa0;µg/mL against Hela and MCF-7 cell lines, respectively. Conclusion: <i>Colletotrichum siamense</i> can be used to develop a sustainable bioprocess production of vinblastine as an alternative to plant extraction.</p>
