Anti-obesity drugs: comprehensive review and emerging therapies

Obesity, an epidemic of modern world lifestyle, has a substantial negative impact on both health and the economy. Traditional weight management strategies, such as dietary modifications and lifestyle changes, are often inadequate for long-term weight control, warranting the immediate intervention of pharmaceutical assistance. This review article evaluates the status of presently approved anti-obesity medications like lipase inhibitors, incretin-based medicines, dual agonists, and melanocortin receptor agonists for their adverse events, alongside novel therapeutic candidates under clinical investigation. The article comprises the impacts of these medications on energy balance, appetite regulation, and metabolic health in the assessment of their mechanisms of action, efficacy, safety profiles and their therapeutic potential with respect to obesity phenotypes. This review article also sheds light on recent promising advancements in anti-obesity innovative therapies such as neuromedin U receptor agonists, amylin/calcitonin dual agonists, and a non-peptide oral GLP-1 receptor agonist. Key future research strategies and directions include the development of multi-targeted and combination therapies were also discussed.
