In-vitro and In-silico Evaluation of Supercritical Extract of Acacia Auriculiformis against Breast Cancer

To study the preliminary effect of the supercritical fluid extract of Acacia auriculiformis (SEAA) against breast cancer. Bark samples of A. auriculiformis were subjected to supercritical-fluid extraction, and the resulting SEAA was analyzed using gas chromatography-mass spectroscopy (GC-MS). Molecular docking was conducted to assess the interaction profile of betulin and doxorubicin with P21-activated kinase 4 (PAK-4), β-cell lymphoma 2 (BCL-2), and Schlafen-12 (SLFN-12). The cytotoxic and anti-proliferative effects of SEAA were examined through the brine shrimp lethality assay and Allium cepa assay. Three doses of SEAA (100 µg/mL, 200 µg/mL, and 300 µg/mL), methotrexate (100 µg/mL) as a standard group, and 0.7% saline as a normal control (NC) group were selected for both assays. The effect of SEAA on cell viability in tumor cells was evaluated through MTT assay on MCF-7 and HT-29 cell lines. GC-MS analysis confirmed the presence of betulin at a retention time of 5.662 min. Betulin and doxorubicin demonstrated similar interaction profiles with PAK-4, BCL-2 and SLFN-12. SEAA treatment significantly (P-value < 0.05) reduced nauplii survival and increased relative lethality. SEAA (100 µg/mL) and SEAA (200 µg/mL) treatments significantly (P-value < 0.05) reduced the number and length of roots, while SEAA (300 µg/mL) treatment produced the maximum reduction in the number (P-value < 0.01) and length (P-value < 0.05) of roots comparable with methotrexate (100 µg/mL) treatment. SEAA demonstrated a dose-dependent reduction of cell viability in MCF-7 and HT-29 cell lines. SEAA treatment demonstrated preliminary cancer-protective potential through in-silico and in-vitro assays.
