<p>Natural proteins frequently contain a helical domain known as the CCDC (Coiled-Coil domain containing). CCDC proteins are encoded by about 180 genes, which are essential proteins involved in various physiological and pathological processes. CCDC proteins play an important role in male gametogenesis (sperm flagellum assembly), embryonic patterning (left–right axis, neural-crest migration) and multi-organ homeostasis (heart, brain, skeleton, eye, auditory). Furthermore, CCDC proteins can influence physiological processes at the cellular and organ levels associated signaling pathways or transcription factors. Moreover, the pathophysiology of many disorders is linked to the abnormal expression of the CCDC protein family. In this article, we review the structure and function of CCDC proteins in human development and the potential of CCDC protein dysregulation as a novel target for clinical diagnosis and treatment to provide ideas for its effects on the onset and course of disease.</p>
