Effects of DpTF Gene Overexpression on Dunaliella parva

This study overexpressed DpTF in D. parva to investigate its effects on organic content and antioxidant activity. The transgenic D. parva with recombinant vector pBI221-UbiΩ-CAT-DpTF was D. parva T. The transgenic D. parva with empty vector pBI221-GFP-UbiΩ-CAT was D. parva C. Compared with D. parva C, protein contents of D. parva T decreased by 34.3 and 48.8 mg/g DW at 12 days and 15 days, respectively. Compared with D. parva C, carbohydrate contents of D. parva T decreased by 9 and 6.2 mg/g DW, respectively. Compared with D. parva C, the contents of chlorophyll a, chlorophyll b, and carotenoid decreased by 47.5%, 43.1%, and 43.27% in D. parva T, respectively. Compared with D. parva C, reducing powers of D. parva T decreased by 3.3% and 6.3%/mg at 12 days and 15 days, and hydroxyl radical scavenging activity of D. parva T decreased by 24.8%/mg at 12 days. Oil contents of D. parva T decreased by 3.5% and 14.7% at 12 days and 15 days, compared with D. parva C. It was inferred that DpTF overexpression resulted in the reduced mRNA levels of DpWRI1-like and DpAP2, further reducing oil content and carotenoid content. The results showed that DpTF had inhibitory effects on organic contents and antioxidant activities.
