<p>Populations of <i>Cereus fernambucensis</i> Lem. (Cactaceae) have shown an alarming decline due to habitat loss. The aim of this study was to determine the influence of in situ and ex situ conservation conditions on the physiological quality of stored seeds of <i>C</i>. <i>fernambucensis</i>. The seeds were stored in organza bags and kept in a cold room (12 ± 2&#xa0;°C and 50% RH) in the dark (ex situ conservation) or buried in soil up to 5&#xa0;cm deep in an area where the species naturally occurs (in situ conservation). The germination of seeds from in situ and ex situ conservation was evaluated for periods of 0, 45, 90, 135 and 180 days. The variables assessed were the seed water content (%), germination (%), germination speed index (GSI), mean germination time (MGT), length, dry mass and biomass density of the seedlings. There was a 75% reduction in the germination of seeds kept in situ, and there was an almost total loss of viability at 180 days of storage in the field (germination &lt; 20%). In contrast, the seeds stored in cold storage maintained more than 93% germination throughout the analysis period, demonstrating that ex situ conservation is the most efficient way of maintaining the viability of the seeds of this endemic species of Brazilian <i>restingas</i>. Under natural conditions, <i>C</i>. <i>fernambucensis</i> seeds are capable of forming a potential short-term seed bank.</p>
