<p><i>Enterococcus faecalis</i> can cause life-threatening infections in humans. The aim of the present study was to explore the traceability of antimicrobial resistance (AMR) contamination in fish from point of production to consumption using <i>E. faecalis</i> as an indicator organism. To achieve this, AMR profiles and virulence factors of <i>E. faecalis</i> isolated from freshwater food fish sold in retail markets, and from fish collected directly from fish farms were compared. A total of 175 gill/muscle swabs were collected and processed for bacterial isolation. The study covered 87 freshwater fish farms located in five districts of Uttar Pradesh, India, and 88 samples from six retail fish markets. The results revealed no significant difference in the distribution of <i>E. faecalis</i> between market (46.5%) and farm (51.7%) samples. Overall prevalence of <i>E. faecalis</i> in freshwater food fish was 49.14%<i>.</i> The market isolates showed significantly higher (<i>p</i> &lt; 0.05) resistance to erythromycin, tetracycline and quinopristin-dalfopristrin than the farm isolates. Resistance to chloramphenicol, ciprofloxacin and high-level aminoglycoside resistance was observed only in market isolates. Multidrug resistance was also seen only in market isolates. Higher number of antimicrobial resistant genes were detected in market isolates than farm isolates. Virulence genes, <i>asa</i>1, <i>cyl</i>A and <i>esp</i> were also detected only in the market isolates. These findings indicate that the market isolates of <i>E. faecalis</i> carried higher AMR than farm isolates, likely due to improper food chain logistics. The study highlights the need for improved sanitary measures during the transportation, storage, and distribution of freshwater fish.</p>
