Exploring efficacy of Pione vastifica ethanolic extract upon Aflatoxin B1: detoxification, immune modulatory and antibacterial effectiveness

Aflatoxin B1 (AFB1) is one of the most prevalent and hazardous mycotoxins and has been strongly associated with hepatotoxicity and an increased risk of hepatocellular carcinoma. This study aimed to evaluate the hepatoprotective, immunomodulatory, antibiofilm, and cytotoxic activities of Pione vastifica extract (PVE) against AFB1-induced hepatic injury and multidrug-resistant bacterial strains. PVE exhibited significant antibiofilm activity against all tested bacterial strains. It also showed dose- and time-dependent cytotoxicity against HepG2 hepatoma cells, with IC50 values of 64.9 and 26.65 μg/mL after 24 and 48 h of exposure, respectively. In the in vivo experiment, AFB1 administration induced marked hepatic injury, as evidenced by impaired liver function, increased oxidative stress, inflammatory responses, DNA fragmentation, caspase-3 expression, and histopathological alterations. Post-treatment with PVE for six weeks significantly ameliorated AFB1-induced hepatotoxicity. This protective effect was associated with increased serum albumin and total protein levels, enhanced hepatic antioxidant defenses including GSH, SOD, and CAT, and reduced serum AST, ALT, ALP, GGT, IL-1β, TNF-α, AFP, hepatic MDA, NO, and DNA fragmentation. PVE treatment also attenuated caspase-3 expression and improved the histological architecture of liver tissue.Overall, the findings suggest that PVE may exert protective effects against AFB1-induced hepatic dysfunction through antioxidant, anti-inflammatory, anti-apoptotic, and tissue-restorative mechanisms. In addition, its antibiofilm activity and cytotoxic effect against HepG2 cells indicate potential biomedical applications. Further mechanistic and safety studies are required before considering PVE as a preventive or therapeutic supplement against mycotoxin-related complications.
