<p>Fructooligosaccharides (FOS) present in pitahaya pulp have a potential prebiotic effect since they are selectively fermented by the intestinal microbiota. The aim of this study was to evaluate the extraction conditions, partial purification, and quantification of FOS present in aqueous and ethanolic extracts of pitahaya pulp. Their resistance to hydrolysis was analyzed using the INFOGEST 2.0 system. The effect of the extract as a carbon source substitute to stimulate the bacterial growth of three Lactobacillus strains: <i>Lacticaseibacillus casei</i> DG<sup>®</sup>, <i>Lacticaseibacillus rhamnosus</i> LGG<sup>®</sup>, and <i>Lactobacillus delbrueckii</i> NRRL B445 was studied. FOS were obtained following a 2<sup>2</sup> factorial design with four central points to evaluate the effect of two factors: sample/solvent ratio (A) and type of solvent (B) on the extraction of FOS from freeze-dried pitahaya pulp. After extraction, partial purification was performed by ultrafiltration with a membrane cutoff of 1&#xa0;kDa size. Data analysis determined that the best treatments were a solid/solvent ratio of 1:10 w/v with water and 1:10 w/v with 80% ethanol. The dried extracts contained FOS levels ranging from 77 to 81&#xa0;g/100&#xa0;g, which were enhanced by the ultrafiltration process. An average resistance to hydrolysis of 52.87% was found, as well as the capacity to stimulate the growth of the strains <i>Lacticaseibacillus casei</i> DG<sup>®</sup>, <i>Lacticaseibacillus rhamnosus</i> LLG<sup>®</sup>, and <i>Lactobacillus delbrueckii</i> NRRL B445. Due to these properties, fructooligosaccharides extracted from dragon fruit pulp could have a potential for use as prebiotics.</p>
