Changes in the oxalate oxidase activity during germination of three cultivars of paddy rice (Oryza sativa L.)

Consuming large amounts of food containing oxalates, an anti-nutrient found widely in plant foods, can raise the risk of kidney stone development and lead to mineral deficiencies. Oxalate oxidase (OXO), a degrading-oxalate enzyme, has been reported to be existing in germinated cereals. Isolation of the crude enzyme from germinated rice, a staple food in Asian countries, could be potential to provide a safe treatment for people suffering with kidney stones and other disorders related to oxalates. In this research, the presence of OXO in germinated paddy rice at different germination time had been detected in three cultivars: Oryza sativa L.cv. OM 4900, Oryza sativa L.cv. OM 5451, and Oryza sativa L.cv. IR 4625. The activity was determined using Ultra high-performance liquid chromatography system. Comparison between 48-hr immersed paddy rice and unsoaked seeds showed that there were significantly increases of the activity in the soaked seeds of all studied cultivars. Moreover, the highest OXO activity was detected in the 3-day germinated IR 4625 variety, at 32.90 U/g DW, which had increased 5.8-fold as compared with un-germinated ones. Total protein contents of three cultivars which relate to the specific activity OXO tended to reduce gradually during the seed germination.
