Increased production of vincamine—an important medicinal drug in hairy root culture and mist bioreactor

Vincamine is a monoterpenoid indole alkaloid found in Vinca minor L. Its dextrorotatory configuration, obtained through semi-synthetic processes and compound resolution, is used for treating cardiovascular and cerebrovascular diseases. Vincamine from V. minor also exhibits the dextrorotatory form, but its low abundance makes chemical synthesis the primary production method for pharmaceutical applications. In this study, a gene involved in vincamine biosynthesis was cloned from V. minor leaves and integrated into a Ri plasmid. The positive hairy roots lines were obtained by genetic transformation, which achieved a 4-fold increase in vincamine content. Furthermore, a preliminary mist-type bioreactor cultivation system was established, resulting in an average growth rate of 0.17 g day−1 of hairy roots after 30 days (d) culture.
