The potential of banana extract and light quality using LEDs to optimize in vitro growth of Paphiopedilum glaucophyllum J.J. Sm.

The slipper orchid Paphiopedilum glaucophyllum J.J. Sm. is an endemic species to Java Island, Indonesia, which is currently facing extinction. This research aimed to enhance the growth of P. glaucophyllum with modifications of growth media and growth maintained under various wavelengths using light-emitting diodes (LEDs). Two-year-old plantlets were cultured in Murashige and Skoog (MS) medium supplemented with banana extract (0, 25.0, 50.0, 75.0, and 100.0 g L−1). The plantlets were also cultured in MS medium with 5.0 mg L−1 kinetin and 1.0 mg L−1 IAA as a positive control. The plantlets were exposed to white, red, blue, and red-blue LEDs for 90 d. The new shoots of P. glaucophyllum emerged between 50.6 and 63.0 d after subculturing, with the fastest shoot emergence and the most optimal number of shoots occurring in a combination of 25.0 g L−1 banana extract and blue LED, respectively. A combination of 100.0 g L−1 banana extract and blue LED light resulted in optimal growth of plantlet height (1.27 cm ± 0.07). The positive control showed the highest number of leaves (2.66 ± 0.23) under white LED light, while a significant increase in leaf length (0.79 ± 0.05) was recorded under red LED light. All treatments produced an optimal number of roots under red LED light except for the negative control. Furthermore, the 50.0 g L−1 banana extract produced the highest chlorophyll content under red LED light. These findings suggested that further research into the effects of banana extract and light quality on the growth of Paphiopedilum is a promising avenue for species conservation.
