Effects of elicitor treatments and harvesting times on rosmarinic acid production in cell suspension culture of Satureja khuzistanica

The efficiency of different biotic and abiotic elicitors was investigated on the RA production in the cell cultures of Satureja khuzistanica. MeJ (0.05, 0.1, 0.2, and 0.5 mM), YE (1.0 and 5.0 mg.L −1), Cor (1.0 and 2.0 mM), Van (0.074, 0.153, and 0.306 mM), and SA (0.02, 0.1, and 1.0 mM) elicitors were applied at 3 and 11 days (d) after culture establishment, and RA content of the treated cells were measured at 6, 24, 48, 72, 96, 168, and 264 h after treatment. Both time of elicitor treatment and cell harvesting significantly affect the RA content of cultures. The highest RA accumulation was attained when elicitors were applied 11 d after cell culture establishment. The optimum time for cell harvesting varied among the elicitors, as the maximum RA yield in MeJ, YE, Cor, and Van was recorded when cells were harvested 48 h after elicitor treatment, while SA-treated cells achieved the highest RA 72 h after treatment. Among the elicitors, MeJ (0.05 mM) and SA (1.0 mM) showed to be most efficient for inducing RA biosynthesis in S. khuzistanica. However, the highest RA yield (3294.23 mg.L−1) was obtained in the 0.074 mM Van-treated cells, followed by 0.05 mM MeJ (3036.04 mg.L−1). Elicitor treatment is recommended 11 d following the establishment of cultures, with cell harvesting occurring 48 to 72 h post-treatment, to maximize RA production in S. khuzistanica. These findings highlight the significance of optimal times for elicitor treatment and cell harvesting to improve RA yield in cell suspension systems.
