<p><i>Juglans mandshurica&#xa0;</i><i>Maxim</i>, a valuable timber and economic species in Northeast China, faces challenges in seed propagation due to its dormancy. Somatic embryogenesis is one of the important pathways for forest tree propagation and genetic improvement. <i>WOX8</i> is essential for maintenance cell proliferation during both embryonic and post-embryonic development. In this study, <i>JmWOX8</i> gene was cloned, containing a 906-bp CDS sequence encoding 301 amino acids. Subcellular localization confirmed its nuclear localization. The amino acid sequence of JmWOX8 contains a conserved homeobox domain belonging to the Homeodomain superfamily, and shares &gt; 70% sequence similarity with WOX8 homologs from other species. The qRT-PCR results revealed <i>JmWOX8</i> expression in both embryogenic callus (EC) and non-embryogenic callus (NEC) followed a unimodal pattern, peaking on the 21st day and then dropping sharply by the 28th day. These results suggest that <i>JmWOX8</i> plays an important role in the early stage of somatic embryogenesis of <i>J. mandshurica.</i> The research provides a foundation for elucidating the molecular mechanisms of somatic embryogenesis of <i>J. mandshurica</i>. </p>
