Micropropagation of elite varieties of marigold (Tagetes erecta L.)

Marigold (Tagetes erecta L.), an asteraceous species with industrial and therapeutic significance, synthesizes essential compounds such as pyrethrins, thiophenes, and lutein, providing insecticidal, nematicidal, and nutritional advantages. This study optimized direct organogenesis in marigold (Tagetes erecta L.) through shoot tip and leaf culture of Pusa Basanti Gainda (PBG) and Pusa Narangi Gainda (PNG) from germinated seedlings, emphasizing the impact of plant growth hormones added to Murashige and Skoog (MS) medium that were tested for their effect on shoot tip multiplication and leaf explant response to direct organogenesis. In both varieties, shoot tip culture on MS medium without growth regulators resulted in 100% shoot induction, whereas higher regulator concentrations led to vitrification. PBG demonstrated superior leaf explant response, with MS medium containing 2.0 mg L−1 BAP (6-benzylaminopurine) and 0.5 mg L−1 NAA (naphthaleneacetic acid) achieving 90.67% micro-shoot formation efficiency and an average of 5.83 shoots per explant. Genetically clonal in vitro regenerated plantlets were confirmed by polymerase chain reaction (PCR) based on molecular analysis using inter simple sequence repeats (ISSRs). This in vitro regenerated protocol may be useful for the large-scale production of identical plantlets and provides a foundation for genetic transformation, genome editing, and breeding programs in marigold.
