<p><i>Musa acuminata</i> AAA Cavendish cv. ‘Grand Nain’ is susceptible to banana bunchy top virus (BBTV) and exhibits a compatible host–pathogen interaction. The BBTV-Raipur isolate identified in native <i>Musa</i> field plants in Chhattisgarh, India, was collected and described. It was identified as a distinct member of the Pacific-Indian Oceans (PIO) group by a phylogenetic analysis of 29 nucleotide sequences of the BBTV DNA-S coat protein gene. Further, <i>in vitro</i>, an investigation was conducted to understand the altered physiology of the BBTV-infected regenerants in comparison to the healthy <i>Musa</i> regenerants (control) by studying the growth attributes and biochemical stress indicators such as the amount of leaf pigments, hydrogen peroxide (H<sub>2</sub>O<sub>2</sub>) content, catalase (CAT), and ascorbate peroxidase (APX) activity were determined. Morphological parameters such as shoot proliferation, pseudostem height, girth, and leaf area were reduced significantly in the BBTV-infected regenerants, in contrast to the control. Also, the photosynthetic pigment Chl <i>a</i> content was reduced significantly. With the higher accumulation of the oxidative metabolite H<sub>2</sub>O<sub>2</sub>, APX activity was significantly higher, while the activity of the CAT declined in the BBTV-infected regenerants. Thus, the present findings indicated that the presence of the BBTV particles in the <i>Musa</i> host cells leads to accumulation of H<sub>2</sub>O<sub>2</sub> resulting in declined CAT activity and disturbance in the chloroplast, while the APX activity enhances targeting the accumulated H<sub>2</sub>O<sub>2</sub>. Hence, as a consequence of alteration in the functioning of the cells and disruption of normal physiology, <i>in vitro</i> BBTV-infected <i>M. acuminata</i> regenerants exhibit inferior growth and development.</p>
