In vitro micropropagation of Theriophonum sivaganganum (Ramam. and Sebastine) Bogner—an endemic plant of South Tamil Nadu, India

An efficient in vitro micropropagation protocol was developed for Theriophonum sivaganganum (Ramam. and Sebastine) Bogner, an endemic species to the South Tamil Nadu in India. This is the first report on the genus which has been medicinally used by the local tribes and also potential members of ornamental plants. The present study established a reproducible protocol of T. sivaganganum using corm terminal buds (CTBs) and corm discs (CDs) explants. The sterilized corm explants were pre-treated with L-ascorbic acid (AA) prior to inoculation on Murashige and Skoog (MS) medium augmented with different plant growth regulators (PGRs) for in vitro shoot multiplication. The shoot initiation response was high (87.66%) on CTB explants followed by CD explants (82.10%) on N6-benzyladenine (BA) at 1.0 mg L−1 with an average of 3.5 to 3.8 shoots from both the explants. The synergistic treatments of BA with different auxins showed increased response of shoot induction and shoot numbers. The highest number of 8.90 and 17.92 shoots per explants was observed on BA with 0.5 mg L−1 of 1-naphthaleneacetic acid (NAA). The highest number of roots (9.63) per shoot was recorded on half-strength MS medium augmented with 0.5 mg L−1 indole-3-butyric acid (IBA). All the plantlets were effectively hardened and acclimatized upon being shifted to the greenhouse condition with a survival rate of 85%. This technique can be employed for mass multiplication following phytomedicine studies and commercial production purposes.
