Morphological and phylogenetic revision of Triphragmiopsis (Pucciniales): study of type collections and description of Triphragmiopsidaceae fam. nov.

Triphragmiopsis is a small genus of rust fungi comprising three species in the world, characterized by three-celled pedicellate teliospores with distinctly verrucose surface. Two species, T. jeffersoniae and T. laricina, are distributed in East Asia, but T. isopyri is known only from Europe. For a long time, the genus Triphragmiopsis and the morphologically similar genera Triphragmium and Nyssopsora were subsumed under the family Raveneliaceae. In recent studies, DNA sequence-based phylogenies have shown that the latter two genera belong to different families within Pucciniales (Phragmidiaceae and Nyssopsoraceae, respectively). However, a phylogenetic study of Triphragmiopsis has not yet been conducted, and its taxonomic status remains uncertain. In our study, we performed the first molecular phylogenetic revision of the genus Triphragmiopsis using three ribosomal markers (ITS, LSU and SSU). All three species of the genus were represented in our sampling, including the type specimens of T. jeffersoniae, the generic type for Triphragmiopsis. Based on examination of the LEP Fungal Herbarium located specimens corresponding to the original materials, one specimen was designated as the lectotype of T. jeffersoniae. We performed detailed morphological observations of all studied specimens using both light and scanning electron microscopy. The ITS phylogeny revealed that T. jeffersoniae, T. isopyri and T. laricina clustered together and formed a highly supported monophyletic clade. The position of the Triphragmiopsis clade within the Pucciniales was assessed by phylogenetic analysis of ITS + LSU and LSU + SSU datasets. Based on these analyses, we introduced a new family Triphragmiopsidaceae to accommodate Triphragmiopsis species.
