Depressiostiolaceae fam. nov. (Pleosporales, Dothideomycetes) for reclassification of two Massaria species

Massaria ginkgonis on Ginkgo biloba (Ginkgoaceae) has been placed under Massaria based on its ascospore morphology since the original description. To clarify the phylogenetic position of M. ginkgonis, DNA sequences from five regions of the fungus—the small subunit (SSU) of the nuclear ribosomal DNA, the nuclear ribosomal internal transcribed spacer (ITS) region, the large subunit (LSU) of the nuclear ribosomal DNA, the second largest RNA polymerase II subunit (RPB2) gene, and the translation elongation factor 1-alpha (TEF1) gene—were generated from the strains and used for molecular phylogenetic analyses for the first time. Massaria ginkgonis clustered with Asteromassaria olivaceohirta (synonym: M. olivaceohirta) on Morus spp. (Moraceae) and formed a lineage independent of any known family in Pleosporales. A novel family, Depressiostiolaceae, is proposed to accommodate the new genus Depressiostiolum. Depressiostiolum is characterised by deeply immersed ascomata with a depressed ostiolar neck. Based on the morphological observations and molecular phylogenetic analyses, two new combinations of Depressiostiolum, D. ginkgonis and D. olivaceohirtum, are proposed. Both species may have expanded their distribution with host naturalisation from China, considering their presence in the immigrant areas of host plants.
