First Report of Root-lesion Nematode, Pratylenchus oleae (Nematoda: Pratylenchidae) from Potato (Solonum tuberosum L.) in Türkiye

Root-lesion nematodes (Pratylenchus spp.) (RLN) cause serious damage to vegetables in Türkiye. During a survey of root-lesion nematodes of potato in the potato-growing areas of the Central Anatolia region of Türkiye, a root lesion nematode species was extracted from the infected potato roots in Konya and Niğde provinces. After extraction and pure multiplaction in carrot culture, the species was identified based on morphological, morphometric, and molecular methods. For molecular analysis, D2/D3 expansion segments of 28S rRNA and the internal transcribed spacer (ITS1) region of ribosomal DNA were amplified by PCR and sequenced. As a result of all the methods used for diagnosis, we identified the nematode causing the infection as the root-lesion nematode Pratylenchus oleae. To our knowledge, it is reported for the first time from Türkiye and from a potato plant in the world.
