<p><i>Dichroa febrifuga</i> Lour., a traditional Chinese medicinal herb historically used for malaria treatment, contains the active compound Febrifugine. Through structural modification, Halofuginone was semi-synthesized, retaining antimalarial activity with reduced toxicity. Recent studies reveal Halofuginone’s broad biologic activities, including significant therapeutic potential in autoimmune diseases. By modulating abnormal immune responses, suppressing inflammation, and exerting antifibrotic effects, Halofuginone alleviates symptoms and prevents tissue damage in these conditions. This review comprehensively summarizes advances in Halofuginone research for autoimmune diseases and discusses key challenges in this field.</p> Graphical abstract <p></p>
