Modulation of HSP27 and inflammatory cytokines by different resistance training protocols in postmenopausal women

This study aimed to compare the acute effects of two distinct resistance exercise (RE) protocols on circulating levels of HSP27, phosphorylated HSP27 (pHSP27), IL-6, IL-1β, IL-10, and TNF-α in trained postmenopausal women.
MethodsThirteen resistance-trained postmenopausal women participated in a randomized crossover trial involving two RE protocols: high-load (HL: 90% 1RM, 6 repetitions per set, 3 sets, 3-min rest) and low-load (LL: 50% 1RM, 20 repetitions per set, 3 sets, 1.5-min rest). Each session included seven exercises and was separated by a 7-day washout. Blood samples were collected at baseline (PRE), immediately post-exercise (POST), and 1-h post-exercise (1H).
ResultsBoth protocols were performed near concentric failure (repetitions in reserve: HL = 0.91 ± 0.58; LL = 2.41 ± 1.79). LL elicited higher blood lactate (↑92%, p = 0.027) and training volume (↑86%, p < 0.001). Only HL significantly increased circulating HSP27 levels: + 49% from PRE to POST (p = 0.005) and + 66% from PRE to 1H (p < 0.001). LL did not significantly affect HSP27. IL-6 levels rose post-exercise (p = 0.011), with no significant difference between the protocols (p = 0.239). No significant changes were observed in pHSP27, IL-10, IL-1β, or TNF-α.
ConclusionHigh-load resistance training acutely increases circulating HSP27 in trained postmenopausal women. IL-6 levels rise after RE regardless of load, while pHSP27, IL-10, IL-1β, and TNF-α remain unchanged after a single session.
