<p>Bovine mastitis is a highly prevalent disease that causes significant losses in dairy herds. The objective of this study was to evaluate the seroconversion of immunoglobulin G antibodies after autogenous vaccination against mastitis in crossbred dairy cows. Sixty cows were paired based on similarity for days in milk, parity, somatic cell count and milk production. One cow of each pair was randomly assigned to the vaccinated group (<i>n</i> = 30) and the other cow to the Control group (<i>n</i> = 30). A California Mastitis Test was performed on all cows of the herd, and positive cases had their milk samples submitted to microbiological culture. The selected bacterial species for vaccine composition were <i>Corynebacterium bovis</i>,<i> Escherichia coli</i>,<i> Staphylococcus chromogenes</i>,<i> Streptococcus agalactiae</i>,<i> Streptococcus dysgalactiae</i> and <i>Streptococcus uberis.</i> The first vaccination occurred in March 2022 and the booster 30 days later. Blood samples were taken every 30 days for seroconversion analysis by enzyme-linked immunosorbent assay. On the day of vaccination, no significant difference in IgG levels was found among groups, as expected. On days 30, 60, 90 and 120 after vaccination, there was a significant difference (<i>P</i> &lt; 0.05) in seroconversion between groups, which varied according to each bacterial species tested. In conclusion, autogenous vaccination stimulated the immune system of the cows to produce specific antibodies against the bacterial species present in the vaccine.</p>
