<p>Goatpox is a highly contagious viral disease of goats caused by the goatpox virus (GTPV) of the genus <i>Capripoxvirus</i>, family <i>Poxviridae.</i> The huge economic losses in outbreaks are due to hide damage, production losses and high morbidities associated with the disease. An outbreak of goatpox was investigated in a goat flock from Hisar district of Haryana, India, in April 2022 with an overall morbidity of 21%. GTPV was first identified in pock lesions using a polymerase chain reaction (PCR) assay targeting E10R and RPO132 genes and subsequently identified by isolation in Vero cells and further confirmed by sequencing of the P32 gene of GTPV. Cytopathic effects of rounding, clumping and detachment of cells were observed in Vero cells. The PCR amplification of harvested cells amplified a 955&#xa0;bp specific to the P32 gene and confirmed to be GTPV by sequencing. The phylogenetic analysis revealed nucleotide identity of more than 99% with Indian isolates, signifying a close genetic relationship. Based on the median joining network analysis of P32 gene, 33 haplotypes were identified among the GTPV population. Population structure analysis revealed high haplotype (0.991 ± 0.009) and low nucleotide (0.0041 ± 0.0004) diversities and significant negative values for the neutrality indices indicating purifying selection and rapid population expansion. The findings will help to understand the distribution and dynamics of genetic differences among the GTPV populations. Further detailed studies are warranted to delineate the molecular details of the virus for better understanding the dynamics and molecular epidemiology of pox viruses in the country.</p>
