Large-scale production of date palm plants via somatic embryogenesis

Somatic embryogenesis is considered to be the best in vitro culture approach for the mass propagation of date palms. Maturation and germination of somatic embryos are bottleneck phases that affect plant production in this approach. The present study demonstrates a promising system for enhancing somatic embryogenesis of the date palm cultivar Deglet Noor. From various concentrations of glutamine and folic acid supplemented into Murashige and Skoog (MS) medium, 200 mg/l glutamine and 0.5 mg/l folic acid produced the highest number of mature embryos. MS medium with 0.5 mg/l zeatin and 15 mg/l chitosan yielded the highest number of germinated somatic embryos. Addition of kaolin and diatomite (diatomaceous earth; DE) at different concentrations to rooting medium, as well as foliar and soil application of DE in greenhouse conditions enhanced plant growth. Our study demonstrates that the addition of kaolin and DE is beneficial for the tissue culture propagation of date palms.
