<p>Single-cell sequencing technology offers researchers an unprecedented opportunity to analyze cellular heterogeneity and gene regulation at the single-cell level, enabling precise identification of cell types, reconstruction of developmental trajectories, and exploration of cell-specific regulatory networks. This technology was first applied to animal research and has gradually been refined. It has also been widely used in plant research. However, due to the particularity of plant samples, conventional experimental methods cannot be fully and universally applied to all plant samples. This article introduces various experimental methods of plant single cells, especially the development and application of emerging technologies such as FX-cells. We further compare the advantages and limitations of current experimental approaches and discuss potential optimization strategies for improving experimental efficiency, data quality, and the broader application of plant single-cell omics technologies.At the same time, this review focuses on researchers’ exploration in different organs of plants and summarizes it according to the depth of research: roughly divided into the construction of high-resolution cell maps, the reconstruction of complex growth and development trajectories, and then to the analysis of deep metabolic mechanisms and the study of stress response in adverse conditions. In addition, this article explores the trend of the evolution of single-cell technology from single-omics to multi-omics joint analysis. By integrating multi-dimensional data, researchers have been able to more comprehensively reveal the molecular mechanisms of plant growth and development as well as environmental adaptation, providing a new perspective for precise plant biology research.</p>

Single-cell omics in plants: technological advances and biological insights
