Meta-topolin as an effective cytokinin source to enhance in vitro establishment, shoot multiplication and alleviation of shoot tip necrosis (STN) in kiwiberry (Actinidia arguta) cultivars

This study was aimed to develop an efficient and reliable in vitro propagation protocol in four Kiwiberry (Actinidia arguta) cultivars viz., ‘Ananasnaya’, ‘Dumbarton Oaks’, ‘Ken’s Red’ and ‘Meader Male’, respectively. For this, the optimization of critical parameters such as explant sterilization, in vitro establishment, in vitro shoot proliferation, root regeneration and hardening was done. Besides this, during the establishment and multiplication stage some vital factors associated with the occurrence of shoot tip necrosis (STN) were considered so as to overcome this problem. Overall, the inclusion of meta-topolin in MS medium resulted in successful establishment of explants, mass shoot multiplication and circumvention of STN. The healthy multiplied microshoots were allowed to regenerate the roots via two approaches: in vitro and ex vitro. Subsequently, the plantlets were successfully hardened in a potting mixture composed of cocopeat, vermiculite and perlite (3:1:1). This study demonstrated the potential role of meta-topolin (mT) for in vitro propagation of kiwiberry. Thus, the optimized protocol may be used for large-scale production of quality planting material (QPM) of kiwiberry.
