Spermidine and methyl jasmonate enhance polyphenol content and improve antioxidant, anti-inflammatory and skin regenerative properties in Coffea canephora callus culture

Plant biotechnology continues to face the challenge of meeting the increasing demand for high-value therapeutic products. In this study, we evaluated the influence of the elicitors spermidine (Spd) and methyl jasmonate (MJ) on the production of phytochemicals with antioxidant, anti-inflammatory, and skin-regenerative properties in Coffea canephora callus extracts. Callus was induced from C. canephora leaves and treated with Spd or MJ at varying concentrations. The resulting C. canephora callus extracts (CCCEs) were analyzed for total phenolic content, flavonoids, and protein levels. Antioxidant activity was assessed using ABTS, DPPH, and FRAP assays. Intracellular anti-inflammatory activity was evaluated in LPS-stimulated macrophages by measuring the production of nitric oxide (NO), superoxide anion (O2●−), tumor necrosis factor (TNF-α), and the interleukin-6 (IL-6). Callus of 200 µM MJ-treated group showed a remarkable 13-fold and 15-fold increase in phenolic and flavonoid content, respectively, compared to the control. Antioxidant activity in the MJ-treated groups demonstrated IC₅₀ values that were 7 to 9 times lower than those of the untreated controls, indicating significantly enhanced radical scavenging capacity. Furthermore, all CCCEs promoted fibroblast proliferation and migration, while significantly suppressing the production of pro-inflammatory mediators NO, O2●−, TNF-α and IL-6 in a dose-dependent manner. These findings suggest that elicitation of C. canephora callus cultures with Spd and MJ is an effective strategy for enhancing the biosynthesis of polyphenols with potential applications in antioxidant therapy, anti-inflammatory treatment, and skin regeneration.
