Effect of phenolic compounds (especially hydroxybenzoic acids) on in vitro root formation of Jonagold apple shoots

Phenolic compounds are synthesized by plants and are present in all tissues of higher plants. These compounds play various roles in plant development. During rhizogenesis, the addition of phenolic compounds to the culture medium can influence root formation by either activating or inhibiting it. This study aimed to analyze the effect of exogenous application of phenolic compounds on the in vitro rooting of apple shoots. In this study, the activity of various oxidases is evaluated using enzymatic assays (IAA-oxidase, peroxidases and polyphenol oxidase) and correlated with the concentration of free indol-3-actic acid (IAA) level and the percentage of rooting. The effect of specific compounds depends on their chemical structure, particularly the number of OH-groups and their proximity to the carboxylic functional group: the farther the hydroxyl group is from the carboxyl group, the higher the rooting rate (1.8% for control, 17% for p-coumaric acid, and 39% for ferulic acid). However, no rooting occurs with salicylic acid. In the enzymatic study, polyphenol oxidases (PPOs) exhibited variation, with a greater decrease observed when salicylic acid was used (around a 60% decrease). For these explants, the level of endogenous IAA was higher throughout all culture cycles (3 times higher than the baseline).
