Micropropagation via internodal and petiole explants and phyto- pharmacological studies in Lepidagathis cuspidata Nees., an important medicinal shrub

An efficient micropropagation protocol has been standardized for Lepidagathis cuspidata Nees., a spiny medicinal shrub using internodal and leaf petiole explants on MS medium supplemented with different concentrations (0.5 to 2.0 mg L− 1) of BAP, TDZ, NAA and IAA alone or in combination. The highest frequency (89%) of direct multiple shoots (mean number of 22.6 shoots per explant) from internodal explants were observed on MS medium supplemented with BAP (1.0 mg L− 1) and NAA (0.25 mg L− 1). The optimum frequency of 88% indirect shoot induction from petiole derived calli with a mean number of 21.4 shoots per explant was observed on MS medium supplemented with1.0 mg L− 1 BAP. For rooting of shoots, quarter strength MS medium containing 1.0 mg L− 1 IBA was optimum with 83% response and mean number of 16.7 ± 0.42 roots per shoot. The plantlets were successfully acclimatized and transferred to field conditions with 92% survival rate after 90 days of transplantation. The genetic similarity of mother plant and callus derived daughter plants was confirmed by using SCoT analysis. The free radical scavenging activity of different extracts of in vitro derived L. cuspidata was measured by 2, 2- diphenyl-1-picrylhydrazyl (DPPH) assay. Here, the IC50 value of methanolic extract was found to be the highest (63.53 ± 0.35 µg mL− 1). The highest result of total antioxidant content (134.97 mg AAE g− 1), phenolic content (13.35 mg GAE g− 1) and flavonoid content (17.19 ± 0.16 mg QCE g− 1) of in vitro derived plant extracts were obtained in methanolic extract. This is the first report on micropropagation of L. cuspidata which could aid in the large-scale propagation and conservation of this over exploited taxon highly valuable in therapeutic purposes.
