First regeneration of doubled haploids via anther culture in Triticum timopheevii

Timopheev’s wheat (Triticum timopheevii) is a tetraploid species with unique genetic traits, a good stress tolerance, and a high potential for breeding. However, its utilization in tissue culture is limited due to its poor in vitro regeneration. A method for embryo induction and plant regeneration via anther culture has been described in the breeding line Georgya. The induction medium NPB-99 was the most effective for embryo formation, compared to both C17 and 190-2. The protocol described allows obtaining approx. 1 green plant per 100 cultured anthers. Regardless of the medium composition, T. timopheevii produced a very high percentage of albino plants, reaching 96% on 190-2 medium. Among the green progeny tested by flow cytometry, 42% of plants were spontaneously doubled haploids, 49% were haploids and 9% were hexaploids. This research describes the very first attempt to optimize a protocol for androgenesis induction in T. timopheevii and may serve as a starting point for further development.
