Induction of tetraploids in vitro by three antimitotic agents and their mode of promotive effects on formation of adventitious shoots and phenotypic traits in Campanula medium L.

The effects of antimitotic agents on the tetraploid induction of Campanula medium L. and morphological characters were investigated. Seeds of three cultivars of C. medium, namely, ‘May Purple’, ‘May Pink’, and ‘May Purple Mardine’, were treated with amiprofos-methyl and oryzalin (0, 32.9, and 65.8 µM), and colchicine (1,600, and 3,200 µM) for 3 or 7 d. Colchicine and amiprofos-methyl effectively induced adventitious shoots, whereas oryzalin showed no effect. At 3,200 µM for 7 d, colchicine induced adventitious shoots on the epicotyl and hypocotyl at 59.0% in ‘May Pink’, 61.0% in ‘May Purple Mardine’, and 78.5% in ‘May Purple’. The same treatment induced frequencies of tetraploids at 45.2%, 48.9%, and 50.0%, respectively. Amiprofos-methyl induced no adventitious shoots or tetraploids in ‘May Pink’, but effectively induced shoot formation in ‘May Purple Mardine’ (46.0%) and ‘May Purple’ (48.0%), with corresponding tetraploid induction frequencies of 46.8% and 55.0% respectively, at 65.8 µM for 7 d. ‘May Pink’ showed normal seedling growth with minimal browning, while ‘May Purple Mardine’ and ‘May Purple’ exhibited early browning of the epicotyl and hypocotyl, which preceded adventitious shoot formation. This suggests that browning timing is linked to stress-induced regeneration and reflects genotype-specific sensitivity to amiprofos-methyl. The tetraploid plants exhibited beneficial floricultural traits, including darker flower colors, larger flower and leaf sizes, and increased thickness compared with diploid plants. These findings revealed agent-specific mechanisms of polyploidization and provided a route for generating robustness in C. medium. These tetraploid lines will be noble genetic resources for the ornamental breeding program in C. medium.
