Micro-propagation and mass production of Lophopetalum Wightianum Arn., a globally threatened evergreen tree species in Bangladesh

Lophopetalum wightianum Arn. (Raktan) is a threatened evergreen tree species native to Bangladesh, facing significant propagation challenges due to its recalcitrant seeds and habitat loss. The current study presents the first successful in-vitro micropropagation protocol for Raktan, aimed at developing a reliable method for mass propagation and conservation of this endangered species. Through optimization of culture media and growth regulators, the study achieved about 80% explant survival and successful shoot proliferation using MS medium supplemented with BAP (2.0 mg/L) and Kinetin (1.0 mg/L). In-vitro rooting trials demonstrated recalcitrance, with a 40% rooting rate observed on quarter-strength MS medium containing 3.0 mg/L IBA and 0.5 mg/L IAA after three months. However, ex-vitro rooting through transient dipping showed a higher rooting success rate of 55%, with shoots dipped in MS medium supplemented with 50 mg/L IBA for 72 h. Remarkably, 93% of in-vitro rooted plants survived during the acclimatization phase in field conditions. The findings of this study establish a robust in-vitro micropropagation protocol for the conservation of L. wightianum and provide a foundation for future research and conservation strategies.
