<p>This work presents a newly designed, highly sensitive SPR sensor structure: BK7 Prism/Au/TeO<sub>2</sub>/Bi<sub>2</sub>Te<sub>3</sub>/Sensing medium. Tellurium dioxide (TeO<sub>2</sub>) oxide layer is used to enhance the sensitivity and Bismuth Telluride (Bi<sub>2</sub>Te<sub>3</sub>) as a BRE layer for the attachment of targeted analytes. Here, a high RI BK-7 prism is used to couple incident light with the sensor chip to achieve high sensitivity. The structure is analytically analysed using MATLAB and COMSOL software to plot SPR curves and for field analysis, respectively. The sensor has been meticulously tuned for different gold (Au), TeO<sub>2</sub> layer thicknesses, and Bi<sub>2</sub>Te<sub>3</sub> layer counts to improve sensor performance. The result indicates the achieved sensitivity of (294.12 Deg./RIU), full width at half maximum (FWHM = 0.1182 Deg.), figure of merit (FOM = 34.48 RIU<sup>−1</sup>), and detection accuracy (DA = 0.1186 Deg.<sup>−1</sup>). In comparison to the results reported in the literature, the suggested sensor design performs better and is a good fit for biosensing applications.</p>
