Triple-color PLOP-FISH and Genome Size Analyses of Three Hibiscus syriacus F1 Hybrids and Parental Cultivars

Hibiscus syriacus L. is an ornamental shrub popular in temperate and subtropical regions and various cultivars have been exploited for breeding purposes. Cytogenomic research provides information on chromosome composition, ploidy, and specific sequence distribution useful for breeding. Three H. syriacus F1 hybrids ‘Haeoreum’, ‘Hanbyul’, and ‘Yunsul’ and their parental cultivars ‘Pulkkot’, ‘Wonhwa’, ‘Sondok’, ‘Hansarang’, and ‘Paedal’ were investigated on chromosome constitution, ploidy, rDNA and telomeric repeat distribution, and genome size through fluorescence in situ hybridization (FISH) using pre-labeled oligonucleotide probes (PLOPs) and flow cytometry. The F1 hybrid ‘Haeoreum’ revealed an aneuploid of tetraploid (aneu-tetraploid; 2n = 4x + 2 = 86) with a basic chromosome number of x = 21 from the aneu-tetraploid parents of ‘Pulkkot’ and ‘Wonhwa’. ‘Hanbyul’ was also aneu-tetraploid but with a different chromosome number of 2n = 4x – 10 = 74 from the parents of eu-tetraploid ‘Sondok’ and aneu-tetraploid ‘Hansarang’. In contrast, ‘Yunsul’ was identified as aneu-pentaploid of 2n = 5x + 5 = 110 from aneu-tetraploid ‘Pulkkot’ and aneu-hexaploid ‘Paedal’ parents. In ‘Haeoreum’ and ‘Hanbyul’ rDNA loci showed balanced inheritance from their parents, displaying two 5S and four 45S rDNA loci. But ‘Yunsul’ displayed two 5S and five 45S rDNA loci, indicating unbalancing inheritance of 5S rDNA loci from its tetraploid and hexaploid parents. Genome size estimation of F1 hybrids revealed that the DNA contents of hybrids were found in between the values of their parents, showing a coordinated relationship between chromosome composition and genome size. This result offered valuable insights into the genetic flow between F1 hybrids of H. syriacus cultivars and their parents.
