Influence of Sodium and Silver Salts of Pyrrolopyrazoles on Innate Immunity Reactions In Vivo

Seven compounds were studied in this work: three sodium salts of pyrrolo[3,4-c]pyrazol-3-ones (Ia-c) and four silver salts of pyrrolo[3,4-c]pyrazol-3-ones (IIa-d). Compound Ia was found to enhance the spontaneous and stimulated production of reactive oxygen species by peritoneal leukocytes. Compound Ic also had an activating effect on stimulated luminol-dependent chemiluminescence (LDCL). An assessment of the effect of sodium salts of pyrrolopyrazoles on phagocytosis activity showed a stimulating effect of Ia on the percentage of phagocytosis of peritoneal leukocytes. Silver salts of pyrrolopyrazoles had an opposite effect on the dynamics of the oxidative burst and phagocytic activity. Compound IId was found significantly to inhibit both spontaneous and simulated production of oxygen radicals. In addition, compounds IIa and IIc had a less pronounced inhibitory effect on stimulated LDCL. Compound IId statistically significantly reduced the percentage of phagocytosis of peritoneal phagocytes.
